Berlangsung di stroma kloroplas, area gelap yang dekat dengan membran tilakoid tempat produksi ATP dan NADPH.

Energi untuk reaksi gelap berasal dari ATP dan reduksi NADPH yang diproduksi selama reaksi terang.

Langkah 1: Karbon Fiksasi

RuBP mengikat CO2 dengan bantuan enzim Rubisco, membentuk 3-PGA sebagai produk awal siklus Calvin.

Langkah 2: Reduksi

3-PGA direduksi menjadi G3P menggunakan NADPH dan energi ATP, menghasilkan gula sederhana sementara.

Langkah 3: Regenerasi RuBP

Beberapa molekul G3P dipakai untuk regenerasi RuBP agar siklus dapat berlanjut lagi.
