Blood agar is an enriched medium used to detect the hemolytic properties of bacteria (like Streptococcus species). It contains 5–10% mammalian blood (usually sheep or horse).

MacConkey agar contains lactose and a neutral red indicator. When bacteria ferment lactose, they produce acid, which drops the pH and turns the colonies pink.

Cystine-Lactose-Electrolyte-Deficient (CLED) agar is the gold standard for urine cultures because it supports the growth of most urinary pathogens and prevents the swarming of Proteus species.

Proteus species are highly motile and create a "swarming" film across the surface of moist media like Blood Agar or Chocolate Agar. Media like MacConkey (due to bile salts) and CLED (due to electrolyte deficiency) are designed to inhibit this.

To perform the Kirby-Bauer disk diffusion method, a lawn culture (or flooded culture) is created to ensure even, confluent growth across the entire surface of the Mueller-Hinton agar.

Alpha: Partial hemolysis (greenish discoloration). Beta: Complete hemolysis (clear zone). Gamma: No hemolysis (no change in the medium).

Bacteria that cannot ferment lactose (like Salmonella or Shigella) do not produce acid; therefore, the colonies remain the natural pale/colorless color of the medium.

E. coli is a classic "Lactose Fermenter" (LF). Salmonella, Shigella, and Proteus are all Non-Lactose Fermenters (NLF).

Xylose Lysine Deoxycholate (XLD) agar is used specifically for the isolation of enteric pathogens from stool samples. It helps differentiate Salmonella (which usually produces black-centered colonies due to H2S production) from Shigella.

Robertson’s Cooked Meat (RCM) broth is used for the cultivation of anaerobic bacteria. The chopped meat particles contain glutathione and unsaturated fatty acids which take up oxygen, creating an anaerobic environment at the bottom of the tube.
